逻辑的规划与构建

      在一篇完整的论文中,完整、清晰的逻辑框架是十分重要的。而这一点又是通过Figure所具体展现。不同的实验结果展示成不同的Sub. Figure,阐述共同问题的、核心的Sub. Figures组合成不同的Figure,Figures间按照清晰的逻辑思路构建成一篇完整的文章。

      所以,在论文中应该包含两层意义的逻辑结构:

      1 论据间的逻辑结构——Figure中各个Sub. Figure的规划;

      2 论点间的逻辑结构——文章中各个Figure的规划。

      对于论据间的逻辑结构,这就要求我们选取具有代表性的结果,通过不同的角度(如,基因、蛋白、细胞、组织、活体等),层次递进,逐步拓展,探讨问题。

      而对于论点间的逻辑结构,除了上述的实验研究角度,还可以根据自身研究的通路出发,对文章的核心问题展开描述。

      Abstract: Aberrant expression of apurinic-apyrimidinic endonuclease–1 (APEX1) has been reported in numerous human solid tumors and is positively correlated with cancer progression; however, the role of APEX1 in tumor progression is poorly defined. Here, we show that APEX1 contributes to aggressive colon cancer behavior and functions as an upstream activator in the Jagged1/Notch signaling pathway. APEX1 overexpression or knockdown in human colon cancer cell lines induced profound changes in malignant properties such as cell proliferation, anchorage-independent growth, migration, invasion, and angiogenesis in vitro and in tumor formation and metastasis in mouse xenograft models. These oncogenic effects of APEX1 were mediated by the upregulation of Jagged1, a major Notch ligand. Furthermore, APEX1 expression was associated with Jagged1 in various colon cancer cell lines and in tissues from colon cancer patients. This finding identifies APEX1 as a positive regulator of Jagged1/Notch activity and suggests that it is a potential therapeutic target in colon cancers that exhibit high levels of Jagged1/Notch signaling.
